Both men were airlifted from Whyalla hospital to Royal Adelaide Hospital.
One female victim was transferred in a private vehicle and treated at Whyalla hospital for minor injuries.
A fourth person was treated at the crash site after suffering an asthma attack.
Eyre Highway was closed for a number of hours as police and major crash investigators scoured the crash site.
Police are now considering driver inattention and high speeds as possible causes for the horrific crash.
Eyre and Western Local Service Area Operations inspector Andrew Thiele said the crash was reported to police at 10am on Friday.
“Police subsequently attended and discovered a grim scene,” he said.
“This crash affects family, it affects emergency services who attend, and it affects the community in general.”
Inspector Thiele said the crash occurred on a straight stretch of road and appealed to drivers to take appropriate precautions when driving long distances.
“Please slow down,” he said.
“Speed and inattention are killers on our roads, if you’re tired please pull over and rest.”
